    Inorganic phosphorus analysis,seeds GUS detection,leaves PCR analysis,several mineral nutritional components contents analysis and simple genetic analysis have been carried out on the two low phytic acid mutant lines and two high-iron transgenic rice lines obtained previously.The results indicated that both low phytic acid mutant lines and high-iron lines showed the great potential application.The anther culture of F_1 hybrid progenies between low phytic acid mutant lines and high iron transgenic rice lines was carried out,and the regeneration efficacy and several seedling traits including ploidy,survival rate and seed-setting were evaluated.The results indicated that the low phytic acid gene and ferritin gene had a slight impact on the process of anther culture.By screening numerous progenies,31 homozygous lines of co-expressing low phytic acid gene and ferritin(Fer)gene were obtained.Further analysis showed that 31 homozygous lines had significantly high iron contents,indicating that they were elite low-phytic acid and high-iron breeding germplasm.

    In order to elucidate the development of embryonic characteristics of giant embryo rice TgeB(Oryza sativa L.)and its wide type TB,the whole development of embryonic characteristics of giant embryo and wide type were anatomical observed and studied.The main results were as follows:(1)The greatest growing speed of embryo of ge-rice appeared during the 6th to 9th day after flowering,and the growing speed of ge-rice was higher than that of the wild type.(2)The embryo size of ge-rice was larger than that of CK at all stages,the embryo length and the embryo thicker of ge-rice were 1.21-1.37 times and 1.37-1.70 times than those of CK respectively;the embryo shoot width of ge-rice were 1.12-1.31 times than that of CK.But there was no significant differences in embryo shoot length between ge-rice and CK at all stages;The embryo radicle length of ge-rice were 1.14-1.22 times higher than that of CK,and the radicle width of ge-rice and CK were almost the same at most developing stages.(3)The sizes of scutellum of ge-rice were larger than CK at all stages,and the cell's length and width of scutellum of ge-rice were about 2.09 times and 1.90 times than those of CK,respectively.(4)The number of cells in embryo of ge-rice was almost no difference compared to that of wide type embryo.The enlargement of embryo of giant embryo rice is exclusive due to the enlargement of scutellum cells.

    In order to identify the haploidy of hybrid progeny through wheat×maize,phenotype and chromosome number of hybrid embryo and plants produced by embryo rescue were analyzed on the method of combining morphology and cytology identification.The results showed that:there were no endosperm in the hybrid ovary,sometimes one dissociative globular young embryo could be found in few ovaries,which maybe grew into wheat plants,but their anthers were obsolete or pollens are yeld in the blooming period,which could be looked as a kind of morphology mark.In this study,the chromosome number of all hybrid progeny plants through wheat×maize were 2n=21,there was no homologous chromosome in whole genomes,and there was no maize chromosomes,also.On the basis of all above results and combining the character of chromosomal base number in Triticum L.plant is x=7,the hybrid progeny plants were identified as true wheat haploid(allotriploids),which was the most important cytology mark to identify haploid through wheat and maize cross.The importance of haploid identification in wheat × maize and the differences of previous relative results were also discussed in this paper.

    The soybean mutant library was created by treating the seeds of soybean "Nannong 94-16" with NaN_3-60Co γ-rays irradiation combination treatment and EMS,inspectively.In M_3 generations,54 mutants of leaf,stem,flower,seed and cotyledon mutants were obtained by treatment of NaN_3-60Co γ-rays irradiation,and 66 mutants by EMS,inspectively.Water-logging tolerance mutants of 14 and the light green leaf color mutant were identified by 644 SSR primer pairs.The results showed that,among 14 water-logging tolerance mutants,more than 100 primer pairs displayed polymorphism in 2 mutants and one to seven SSR primer pairs displayed polymorphism in other plants;for the light green leaf color mutant,39 primer pairs displayed polymorphism.Genetic analysis to the light green leaf color mutant showed that this character was controlled by one pair of recessive nuclear genes.The mutant library created for "Nannong 94-16" will be helpful for the research in soybean functional genomics.

    Based on the established regeneration system of Nanlin 95(Populus deltoids Bartr.cv.Lux×P.euramericana(Dode)Guineir cv.I-45/51),through orthogonal design,an efficient transformation and screening system of Nanlin 95 mediated by Agrobateriurn was developed.The leaves from tissue culture served as excellent explants for transformation,and the factors affecting the transformation frequency,such as preculture time of explants,concentration of Agrobacterium and acetosyringone(AS),infection time and co-infection time were discussed by GUS coloration analysis,and the concentrations of antibiotics to inhibit the growth of Agrobacterium were also discussed.The data were analyzed by software DPS,a suitable genetic transformation system was established.The results indicated that 7 days preculturation,20 min Agrobacterium(OD_600=0.6)infection with 200 μmol/L AS,and 3 days' co-infection would lead to an increase in transformation efficiency,and the medium with kanamycin 50 mg/L was found to be the most optimal for transgenic plantlets selection;stable integration of GUS gene in these plants were confirmed by GUS coloration analysis and PCR test.

    It's widespread existence of codon usage bias in organism which has an important influence for expression intensity of exogenous gene.In this study,coding sequences of 7402 proteins were analyzed by CodonW and CUSP programs in maize genome,calculating frequency of codon usage which encoded amino acids and comparing with variety classes of mode organisms such as:human being,yeast,E.coli,Drosophila.These results had shown that codon bias of maize was difference in various degrees with these mode organisms.Comparing codon usage frequency of maize with dicotyledons and monocotyledons including Arabidopsis thaliana,tobacco,rice,wheat,the consequence indicated that codon usage frequency of maize was obviously different from Arabidopsis thaliana and tobacco which belonged to dicotyledon,but had a consistency with codon bias of rice and wheat which are monocotyl.These analytic results had important guidance for maize gene-transforming and selecting the high-efficient expression system.

    Seedlings of sixteen mat rush varieties planted in the field,and shoots derived from tissue culture of four varieties were used to investigate the effects of modified CTAB method and Isolation Kit method on the extraction quality of genomic DNA.The results showed that A_260nm and OD values of DNA solution of same material extracted by modified CTAB method were lower than those by isolation kit method.The concentrations of DNA solution were 0.65-4.90 μg/ml with the former method and 5.10-32.80 μg/ml with the latter method.Therefore,it was demonstrated that DNA Isolation Kit method was more suitable for genomic DNA extraction of mat rush.In addition,it was also found that extraction material had the significant impact on the extraction quality of genomic DNA,and the shoots developed by tissue culture were better than seedlings planted in the field.

    The effects of TDZ,IAA,NAA and medium on bud initiation of cotyledon of immature embryo from jujube cultivar Jinsixiaozao were evaluated by L_9(34)orthogonal design.The result showed that TDZ was the most important factor for bud initiation followed by medium,IAA,and NAA.The optimum combination medium was on 1/2MS medium containing TDZ(0.6mg/L),IAA(0.5mg/L)and NAA0.3(mg/L)by variance analysis,on which cultured in dark 20d,callus inducing rate was 100% and adventitious bud inducing rate was 41.86%.The optimum media for adventitious bud propagation and for root inducing are on MS medium adding 6-BA 1.0mg/L,IBA 0.1mg/L and on 1/2MS medium supplemented with IBA 1.0mg/L respectively.

    Rooting characteristics of cuttings of 12 Camellia species and cultivars irradiated by 60Co γ-rays was studied in this article.The results indicated that survival rate of cutting of irradiated Camellia increased at the radiation dose of 5,10 or 15 Gy and part of species and cultivars was even higher then control.Survival rate of irradiated Camellia cuttings declined significantly and lower than that of control at irradiation dose of 20 and 25 Gy.Length and number characteristics of cuttings of irradiated Camellia was different,the index increased first,and then decreased,with increasing radiation dose.The suitable radiation dose of Camellia was in the range of 5-15 Gy.the maxium values of average number,length and the longest length of cutting rootage were observed within the range.

    Several factors,such as hormone combination,Acetosyringone(AS),Proline(Pro),co-culture time,pre-culture time and infection time,which affect Agrobacterium tumefaciens mediated gene transformation of Malus baccata Borkh,were studied by comparing rate of the kanamycin resistant shoot.The results showed that pre-culture of the leaves was not beneficial to transformation.There was no evident to improve transformation by adding AS and Pro into bacterial suspension.The highest rate of the resistant shoot was obtained under the condition of infection for 10 min and co-culture for 3d.The optimal hormone combination of leaf regeneration medium after transformation was NAA 1.0mg/L+BA 2 mg/L +TDZ 4 mg/L.The PCR analysis showed that the target gene was integrated into the genome of Malus baccata Borkh.

    dctB gene of Pseudomonas stutzerien codes a two component system sensor named DctB.DctB is a sensor of C_4-dicarboxylate,which could activite transcription factor DctD through phosphorylation.DctB were overexpressed in E.coli BL21(DE3)in this study.The fragment of dctB gene was ligased into vector pET28a and then transferred into E.coli BL21.The maximum expression amount of DctB was detected after 8h induced.

    In order to obtain usefull sweet potato mutants,the seeds of Qinshu 1 were intermittently soaked in the mixture of 0.05% colchicine and 2% dimethyl sulphoxide(DMSO)for 12,24,48 and 72h,respectively.Then,the treated seeds under suitable soaking time were cultured in temperatures of 5℃-6℃,10℃-12℃,15℃-16℃,18℃-20℃ and 30℃,respectively.The results showed that the soaking time of 24h and culture temperature of 15-16℃ gave the best mutation efficiency.Cytological observation was carried out in the progenies and chromosome number variation was not found.Among short vine mutants,Duanwan 3 was selected,which possessed high yield,high starch content,high protein content and high nutritional elements such as Fe,Zn and Mn.

    Dry seed of two equal group LongMu No.803 alfalfa,one was irradiated by mixed high-energy particle field(CR)with five doses,i.e.,109Gy,145Gy,195Gy,284Gy,560Gy,which was generated from E2 beam lines of LINAC of Beijing Electron Positron Collider;the other was irradiated with the same five dosages by the 60Co γ-rays.We study about the damage effects.of these two different irradiation ways by planted the seeds and measured the plant hight,plany weight and the plant qulity.The result shows although CR irradiated alfalfa dry seeds have better result than the 60Co γ-rays in the plant height and weight,but 60Co γ-rays did better in change the alfalfa plant quality.Higher dosage(284-559.8Gy)had better effect on change the CP's content than lower dosage.But high dosage's γ-rays will restrain the EE's content.These two different irradiation ways has visible differences.

    The seed of rapeseed(Brassica napus L.)was carried by satellite "Shijian 8".The cytological effects of space mutation on the root-tip cells was studied in SP_1 generation.The results showed that the mitotic index,rate of chromosome aberration and rate of micronuclei were obviously increased in most of treated materials compared with the control,which indicated that true cytological mutations were induced by space flight.The mutagenic effects were observed higher in the maintainer lines than in the male sterile lines.

    Inter-simple sequence repeat(ISSR)is an effective molecular marker developed on the basis of microsatellite or simple sequence repeat(SSR).In this study 21 ISSR primers,which was screened from 100 ISSR primers,were used to analyze and identify the relationship of 8 cultivars of Boehmeria nivea.PCR amplification showed that 86 clear and repeatable bands were obtained,in which 71 bands appeared to be polymorphic(82.5%).Genetic distance and cluster analysis indicated that the genetic relationships between hybrids and their parents were not equal,the male parent transfer more genetic loci to their descendants,which indicated that hybrids inherited more genetic information from the male parent.In addition,a specific DNA segment was amplified only in the fertile cultivars using the primer U835,and the ISSR-marker was converted into SCAR marker.Which could be used as a male sterile molecular marker-assisted selection of Boehmeria nivea breeding.

    The space effects on characteristics of Scutellaria baicalensis Georgi SP_1,including the field survival ability,plant type traits,reproductive characteristics and peroxidase isozyme.The results showed that the growth of different varieties of Scutellaria baicalensis Georgi was inhibited significantly and variation range of characteristics was widen by space-carrying,meanwhile,the peroxidase isozyme analysis indicated that polymorphism of enzymogram was improved also.

    A strain,which can use methylparathion,and its degradation substrate ρ-nitrophenol as source of carbon,was isolated from polluted water pool of Huayang Pesticide Plant.The strain was identified as Arthrobacter sp.and named L-W.L-W's degradability was analysied at different pH and temperature conditions by using gas chromatography and spectrophotometer.The results showed that the removal rate of L-W reached to 85% of original when the concentration of melthylparathion was 50mg/L;and 99% when the concentration of ρ-nitrophenol was 50mg/L within 7h,respectively.

    Low dose hyperradiation sensitivity(HRS)and induced radioresistance(IRR)are two important phenoma in radiobiology.Illustrating the mechanism of HRS/IRR is important to radiation protection,tumor control and creation new functional materials.Radiation-induced DNA double-strand breaks(DSBs)is primer lesions in radiation-induced inactivation,and failure repair of(or misrepair of)DSBs is a prime cause of cell death.Low-dose hyperradiation sensitivity is more prominent in G2 phase cells compared with cells in G1 or S phase.The mechanism regulating the transition between HRS and IRR is likely to be due to an active process occurring in a small fraction of G2-phase cells.DNA repair plays an important role during HRS/IRR.When the radiation dose was lower than 20 cGy,radiation led to less efficient damage response(HRS).When the dose was higher than 20cGy,DNA double strands broke.ATM and G2 phase specific checkpoint were activated.Cell cycle was arrested.DNA repair and cell survival were promoted,IRR appeared at this time.Although lots of results had been got,many problems were still remained.For example,why some cell lines had HRS/IRR,while others not;whether there was mutated HRS/IRR corresponding to single dose or not.These problems are likely to be the future research directions.

    60Co γ-rays was used to initiate acrylic acid grafting of potato starch to produce super absorbent.The influence of adjuvant and synthesis technology on the performance of super absorbent resin were investigated.The drying super absorbent resin,was produced with assistant X and proper synthesis technology.Which could absorb distilled water 350g/g,tap water 157g/g,0.9% sodium chloride 31g/g,respectively.

    The effects of antioxidants on lipid oxidation of vacuum packaged irradiated cooked streaky pork were studied.The cooked streaky pork were added with 0.02% TeaPolyphenols(TP),rosemary,Tertiary butylhydroquinone(TBHQ),Butylated hydroxytoluene(BHT)and vitamin E separately,then were irradiated with 6 kGy,and stored at 4℃.The results showed that antioxidants can reduce the value of TBA,POV value of irradiated cooked streaky pork,and the effects of TBHQ and TP were better,than other antiatidins.

    White dried tofu as raw materials of Yangzhou dishes bean curd shreds,its shelf life was shorter for the traditional craft-making.This paper through improved its production technology and products for vacuum packaging with radiation durability,the shelf life at 4℃for 3 months was achieved.This technology may also apply to other related products.

    Deinococcus radiodurans has a strong ability to withstand high doses of radiation,which makes it as an ideal candidate for bioremediation of sites contaminated with radionuclides and toxic chemicals.However,no data is available on whether D.radiodurans has a specific sorption capacity to radiocesium for bioremediation purpose.The radiocesium biosorption capacity of live cells of D.radiodurans in the presence of other interfering cations was investigated.The maximum biosorption capacity of radiocesium by D.radiodurans in equilibrium state was about 2,100 kBq/kg(fresh weight basis).Among the tested monovalent cations,NH+_4 had the strongest antagonism on 134Cs biosorption for D.radiodurans.However,this antagonism could only be observed at a concentration as high as 100 mmol/L.Divalent cations,such as Ca2+ and Pb2+ could reduced the biosorption of radiocesium by D.radiodurans.Al3+ and Cr3+ were cytotoxic to D.radiodurans cells,the growth of D.radiodurans cells was inhibited when the concentrations of these cations were greater than 1 mmol/L.

    Different plants on slope were collected and 7Be activity in plants was detected.The results showed that with the growth of plants,7Be activity in plants was increasing;7Be activity intercepted and absorbed by plants on slope per square meter was also increasing and reached the maximum in autumn.The percentage of 7Be intercepted and absorbed by plants was 3.35%-8.44%,which indicated that the 7Be interception-absorption by plants obviously impacted on the 7Be activities in soil.The data also showed that daily average of 7Be increment was variable in different sampling intervals with a variation coefficient of 0.69,but the variation was relatively small in the first four sampling intervals from spring to summer.The relationship between daily average 7Be increment and daily average rainfall in different sampling interval was also analyzed and showed of a positive correlation with a correlation coefficient of 0.64.

    Effects of chlorocholine chloride(CCC)(0,1.5,2.0 and 2.5g/L)sprayed at early growth period,then the absorption of phosphate nutrition and the average yield per hill of potato cv.Zhongshu 3 were studied with 32P tracer technology.The results were showed that CCC increased the phosphate absorption during the first several days after emergence,and the absorbed phosphate was mainly distributed in stems and leaves,and the relative concentration of phosphate in organs was stem>leaf>root>tuber in turn.The total amount of phosphate absorped by plants increased with the growth of plants,but decreased at the late growth period.The total amount of phosphate absorped by plants was the highest when sprayed 2.0g/L CCC at different days after emergence.The average yield per hill had improved by 6.83%,10.01% and 3.08% after sprayed 1.5,2.0 and 2.5g/L CCC,respectively,and was positively correlated at 1% level with the phosphate content of leaves at 36d after emergence.

    An insecticidal compound 6β-propoxycelangulin V was synthesized from celangulin V and 1-propanol through anhydrous iodination and Williamson reactions.The purpose of the study was to develop the microsynthetic method of 6β-propoxycelangulin V.The powdered root bark of Chinese bittersweet(Celastrus angulatus Max.)was extracted by benzene under reflux,followed by purification by macroporous-resin and silica-gel chromatography.The desired product was purified by RP-HPLC and identified by HRMS,1H NMR and 13C NMR.The total yield was about 17%,and the purity was higher than 98%.This method could be applied in radiosythesis of 6β-([1-14C]propoxy)celangulin V.

    Sixteen Simmental heifers(average BW 380kg,aged 1.5),which consumed a corn straw diet,were divided into four groups and supplemented with four levels(0,8,16 and 24mg Cu per kg DM)of copper sulfate to evaluate the effects on reproductive hormone excretion during the oestrous cycle.The results showed that serum estradiol,luteinizing-hormone,follicle-stimulating hormone and progesterone concentrations were significantly increased(P<0.05)with supplement at 8mg/kg copper sulfate of DM.Serum estradiol,estriol,luteinizing-hormone,follicle-stimulating hormone and progesterone concentrations were increased with copper supplement at 16mg/kg of DM,but no significant different with control(P>0.05).This indicated that copper sulfate supplement had positive effects on reproductive hormone during estrous cycle at 8mg/kg of DM.Considering the copper content of basal diet,the optimum recommanded copper level of diet is 13.87mg/kg DM.

    Sixteen Simmental steers(420kg and 2.5 years old)were stratified by weight and randomly assigned in a block design to treatments.Treatments consisted of 0(control),0.3,0.6 or 0.9mg/kg of supplemental Se(selenium methionine)diet DM to study the effects of selenium methionine on nutrient metabolic rate and serum biochemical parameters.The digestibility of organic matter,crude protein,ether extract,neutral detergent fiber and acid detergent fiber in groups supplemented Se 0.3 and 0.6mg/kg were higher than control group(P<0.05).Digestion nitrogen,retention nitrogen and the utilization of nitrogen in groups supplemented of Se,0.6 and 0.9mg/kg were higher than control group(P<0.05).In group of supplemented 0.6mg/kg Se,the apparent deposit rate of Ca,P,Se,Cu,Mo and Mn increased(P<0.05),however S and Zn apparent deposit rate was not affected(P>0.05).Concentrations of serum triglycerides,glucose,albumin,total protein were higher than those of control groups(P<0.05).The activities of serum GPT,GOT,GSH-px and the concentration of GSH in group supplemented 0.6mg/kg Se were higher than those of control group(P<0.05).The concentrations of serum MDA in group supplemented Se of 0.6 and 0.9mg/kg decreased(P<0.05).The concentration of GSH in group supplemented 0.6mg Se were higher than control group(P<0.05).Nutrients digestion and utilization improved by supplemented Se of 0.3-0.6mg.The activities of serum enzyme increased and MDA concentration decreased with supplemented 0.6mg/kg.The optimum supplemented Se dose is 0.6mg Se/kg DM.

    Lepidopteran sex pheromones have been used to monitor and control pest populations in field.The trapping efficiency depends upon specific combination of pheromone chip and trapping facility.The effects of different combinations of Chilo suppressalis(Walker)pheromone baits including rubber chip and capillary bait,and trapping facility including water tray and cylinder in the field were compared,the results showed that traditional trapping equipment(rubber bait + water tray trapping)was better than the new cylinder trapping equipment(capillary bait + cylinder trapping)in trapping efficiency,which elicited differences significantly.In the catching of male moths of C.suppressalis of the first and second generation,the trapping facilities had significant differencs on trapping efficiency,while the types of bait had no significant differencs on the efficiency.These data implied that chemotaxis and approaching behavior of male moths to the pheromone baits should be observed in details in order to redesign the facility and improve the trapping efficiency.
